- After 40 cycles of seminested amplification (94°C for 30s, 60°C for 30s, 68°C for 30s), the PCR product was separated by 1.5% agarose gel electrophoresis and the positive product (∼350 to 400 bp) was purified using a Gel/PCR DNA fragment extraction kit (Geneaid).
